- Member of Parliament (MP) for Sabatia (1989 – 2002, 2007-2013)
Elected unopposed in a by-election to take the Sabatia Constituency seat vacated by his deceased father Moses Mudamba Mudavadi. He went on to win the seat in the 1992 and 1997 general elections, and 2007. - Minister for Supplies and Marketing (1989 – 1993)
- Minister for Finance (1993 – 1997)
Youngest Kenyan to hold the portfolio under President Moi. - Minister for Agriculture (1997 – 1999)
- Minister for Transport, Communications and Information (2000 – 2002).
He was appointed by late President Moi towards the end of his tenure as President. - Vice-President of Kenya (2002)
He was appointed Vice-President by President Daniel arap Moi in the twilight of Moi’s administration, serving for a brief period. At the time, he was one of the youngest people to hold that office. - Deputy Prime Minister (2008 – 2013) Following the post-election violence of 2007-2008, a power-sharing deal created the position of Prime Minister (for Raila Odinga) and two Deputy Prime Ministers. Mudavadi was appointed one of the two deputies.
- Minister for Local Government (2008 – 2012)
Concurrently with his role as Deputy Prime Minister, he oversaw the Ministry of Local Government. - Minister for Transport and Infrastructure (2012 – 2013)

Appointed by President William Ruto after the 2022 general election. He also oversees the Ministry of Foreign and Diaspora Affairs.
